For a single game, the DH setting is on the setup screen itself rather than in a separate options panel. After you pick a season, the "2. Choose a Team" box appears. Under the Ballpark dropdown there's a checkbox labeled "No DH (pitcher bats)". Check it and the pitchers bat in that game. Leave it unchecked and both teams use a DH.
That box starts unchecked every time, whatever year you pick. So for a 1995 NL matchup, or any year before the DH, you'll want to check it yourself.
Settings from a season don't carry over to single games. A season's DH option applies only to that season's games. A single game reads only what you pick on the single game setup screens.
